    One nation under God?: a call for secular rhetorical criticism

    2022 Summer.Includes bibliographical references.This dissertation explores the need for secular rhetorical criticism, an approach to rhetorical scholarship that centers questions of power, privilege, and marginalization in relation to ir/religious pluralism. I contend that such an interconnected rhetorical approach to studying religion would be beneficial in creating a more cohesive conversation within rhetorical scholarship on the relationship of religious pluralism and power. Secular rhetorical criticism is fundamentally concerned with the lives, experiences, and voices of the ir/religiously marginalized and recognizes religious nationalism as part of a hegemonic system that privileges religious homogeneity and inhibits religious pluralism. In four chapters, I demonstrate the utility of engaging in secular rhetorical criticism by offering different approaches to analyzing the implementation of the phrase "under God" into the U.S. pledge of allegiance in 1954. While the phrase "under God" in the pledge is largely framed as example unifying "American civil religion" or benign "ceremonial deism," I argue that in 1954 the pledge was transformed into a theistnormative ritual that promoted a Christian nationalist political imaginary while containing Atheists and secularism. In chapter one, I draw on secular rhetorical criticism to urge scholars to be self-reflective of how their own scholarly language and practices maintain religious hegemonies. Specifically, I point to how "under God" as "civil religion" perpetuates the Myth of Religious Tolerance and I offer the conception of theistnormativity as a more critical descriptor for the fusion of belief in God and national identity. In the next chapter, I urge scholars to utilize secular rhetorical criticism as a lens for considering who is contained and negated by theistnormative texts. By analyzing advocates' justification of the new pledge, I demonstrate how religious and political leaders utilized the rhetorical strategy of prophetic dualism to frame the new pledge as a way to contain Atheists and Secularism. In chapter three, I reflect on how scholars engaging in secular rhetorical criticism need to utilize non-traditional methods to analyze the voices of the ir/religiously marginalized. I demonstrate how the gossip method can be used to speculate about how evidence from archived letters and newspapers suggests political leaders knowingly mischaracterized who supported and opposed the change to the pledge. Finally, I urge scholars to utilize secular rhetorical criticism to disrupt the assumption that the contemporary tensions between secularists and Christian nationalists emerged in the 1960s-1970s. By analyzing the political vocabularies of those writing to President Eisenhower and Congress in response to the new pledge in 1954, I demonstrate how supporters viewed the change as a confirmation of a Christian nationalist political imaginary while those who opposed it saw the new pledge as a threat to democracy from the perspective of a secular political imaginary. Using secular rhetorical criticism to guide my analysis of each chapter, I argue that in 1954 the pledge was transformed into a theistnormative ritual that advanced a Christian nationalist political imaginary while containing Atheists and secularism as part of a larger spiritual-industrial complex. This dissertation looks to the history of the 1950s to reflect on how, in 2022, Christian nationalists are establishing a new spiritual-industrial complex. Rhetorical scholars need an approach to studying rhetoric that will challenge and disrupt this undemocratic movement that undermines the values of religious freedom, tolerance, and equality

    The Critical Role of Chemical Pre-Treatment in Ensuring Cryptosporidium Removal by Filtration of High Quality Source Water

    Drinking water utilities reliant on surface water utilize chemically-assisted filtration (CAF) as a key barrier against the passage of protozoan pathogens, like Cryptosporidium spp. oocysts, to treated water. The goal of this work was to enable system-specific and potentially dynamic assessment of oocyst removal by CAF by using zeta potential as a tool for rapid operational feedback. Specifically, this work focused on systems utilizing high quality, low turbidity (typically <1 NTU) source water, with relatively low C. parvum oocyst concentrations and applied full scale coagulant doses (typically <5 mg/L). In these systems, the formation of settleable flocs is not a necessity because source water turbidities are already low and frequently meet treated water criteria. Rather, coagulation is used to enable particle removal through physico-chemical (i.e., chemically -assisted) filtration, as indicated by filter effluent turbidities that may or may not be indicative of optimal particle destabilization and removal by CAF. Accordingly, the identification of “optimal” coagulant doses can be challenging, and becomes even more challenging when process performance is being assessed, such as when Cryptosporidium oocysts are added to filter influents to evaluate their removal by CAF processes. In Phase 1 of this work, the role of oocyst coagulation during CAF performance demonstrations was investigated. It was demonstrated that appropriate coagulation of oocyst seed suspensions is critical to reflecting “well-operated” CAF performance. A protocol for ensuring optimal coagulation of oocyst seed suspensions during such performance demonstrations was developed and demonstrated at pilot-scale. Here, zeta potential was useful in identifying the coagulant doses needed for maximal particle destabilization and removal by CAF. This pilot-scale approach was then validated using lower, environmentally relevant oocyst concentrations (and much longer pilot-scale investigations) during which the entire filtered volume of water was evaluated. Using this protocol, it was demonstrated that a minimum of 3-log oocyst removal could be achieved by CAF (essentially direct filtration) at a variety of operational conditions. During Phase 2 of this work, the protocol developed in Phase 1 was used to evaluate oocyst passage through CAF processes with different filter designs (bed depths, water temperature) at various operational conditions (suboptimal coagulation, filter ripening, end-of-run operation, and hydraulic surges). Here, because of the high quality, low turbidity source water, adequate coagulation was the dominant control for risk, in contrast to many reported investigations in which more deteriorated source water was investigated and operational period within the filter cycle was a more dominant control over oocyst passage through the CAF process. Here, with the exception of suboptimal coagulation conditions, the pilot-scale filters consistently achieved >3-log C. parvum oocyst removal in an essentially directly filtration mode. Thus, this work demonstrated the critical importance of (1) appropriate particle destabilization by coagulation prior to CAF of low turbidity, low DOC source waters, (2) coagulation of oocysts prior to their addition to filter influent streams during CAF performance demonstrations, and (3) zeta potential as a useful tool for ensuring adequate particle destabilization in situations (i.e. treatment of low turbidity, low DOC source waters) in which extensive particle settling is not likely. In doing so, this work further highlights that Cryptosporidium spp. oocyst removal credits of >2.5 log may be warranted for “well-operated” direct filtration processes.

    BackgroundThe Goeckerman regimen remains one of the oldest, most reliable treatment options for patients with moderate to severe psoriasis. Goeckerman therapy currently consists of exposure to ultraviolet B light and application of crude coal tar. The details of the procedure can be confusing and challenging to understand for the first-time patient or provider.ObjectiveTo present a freely available online guide and video on Goeckerman treatment that explains the regimen in a patient-oriented manner.MethodsThe Goeckerman protocol used at the University of California-San Francisco Psoriasis and Skin Treatment Center as well as available information from the literature were reviewed to design a comprehensive guide for patients receiving Goeckerman treatment.ResultsWe created a printable guide and video resource that covers the supplies needed for Goeckerman regimen, the treatment procedure, expected results, how to monitor for adverse events, and discharge planning.ConclusionThis new resource is beneficial for prospective patients planning to undergo Goeckerman treatment, healthcare providers, and trainees who want to learn more about this procedure. Online media and video delivers material in a way that is flexible and often familiar to patients

    BackgroundPUVA treatment is photochemotherapy for psoriasis that combines psoralen with UVA radiation. Although PUVA is a very effective treatment option for psoriasis, there is an absence of patient resources explaining and demonstrating the process of PUVA. Studies have shown that patients who viewed videos explaining the treatment procedures for various medical conditions had a greater understanding of their treatment and were more active participants in their health.ObjectiveTo present a freely available online guide and video on PUVA treatment designed for patient education on PUVA.MethodsThe PUVA treatment protocol used at the University of California-San Francisco Psoriasis and Skin Treatment Center as well as available information from the literature was reviewed to design a comprehensive guide for patients receiving PUVA treatment.ResultsWe created a printable guide and video resource that reviews the benefits and risks of PUVA, discusses the three types of PUVA (hand-foot soak, full body soak, and systemic), demonstrates the PUVA process, and provides practical tips for safe use.ConclusionOnline media and video delivers material in a way that is flexible and often familiar to patients. This new format is beneficial for prospective patients planning to undergo PUVA treatment, health-care providers, and trainees who want to learn more about this treatment

    Excessive Sensitivity to Uncertain Visual Input in L-DOPA-Induced Dyskinesias in Parkinson’s Disease: Further Implications for Cerebellar Involvement

    When faced with visual uncertainty during motor performance, humans rely more on predictive forward models and proprioception and attribute lesser importance to the ambiguous visual feedback. Though disrupted predictive control is typical of patients with cerebellar disease, sensorimotor deficits associated with the involuntary and often unconscious nature of l-DOPA-induced dyskinesias in Parkinson’s disease (PD) suggests dyskinetic subjects may also demonstrate impaired predictive motor control. Methods: We investigated the motor performance of 9 dyskinetic and 10 non-dyskinetic PD subjects on and off l-DOPA, and of 10 age-matched control subjects, during a large-amplitude, overlearned, visually guided tracking task. Ambiguous visual feedback was introduced by adding “jitter” to a moving target that followed a Lissajous pattern. Root mean square (RMS) tracking error was calculated, and ANOVA, robust multivariate linear regression, and linear dynamical system analyses were used to determine the contribution of speed and ambiguity to tracking performance. Results: Increasing target ambiguity and speed contributed significantly more to the RMS error of dyskinetic subjects off medication. l-DOPA improved the RMS tracking performance of both PD groups. At higher speeds, controls and PDs without dyskinesia were able to effectively de-weight ambiguous visual information. Conclusion: PDs’ visually guided motor performance degrades with visual jitter and speed of movement to a greater degree compared to age-matched controls. However, there are fundamental differences in PDs with and without dyskinesia: subjects without dyskinesia are generally slow, and less responsive to dynamic changes in motor task requirements, but in PDs with dyskinesia, there was a trade-off between overall performance and inappropriate reliance on ambiguous visual feedback. This is likely associated with functional changes in posterior parietal–ponto–cerebellar pathways

    The ability to get to where you want to go, when you want to go there is a key factor for aging-in-place in our communities. It is often taken for granted until that ability is compromised. The informal network of family and friends, if it exists, is not likely to be a sustainable transportation alternative for persons with cognitive impairment or for older adults with limitations that may not fit eligibility criteria for senior transportation services, where they exist. The purpose of this study was to investigate the potential of communities to address the specialized supportive mobility needs of community-residing older adults.     Analyzing the ER stress response in ALS patient derived motor neurons identifies druggable neuroprotective targets

    Amyotrophic lateral sclerosis (ALS) is a degenerative motor neuron (MN) disease with severely limited treatment options. Identification of effective treatments has been limited in part by the lack of predictive animal models for complex human disorders. Here, we utilized pharmacologic ER stressors to exacerbate underlying sensitivities conferred by ALS patient genetics in induced pluripotent stem cell (iPSC)-derived motor neurons (MNs). In doing so, we found that thapsigargin and tunicamycin exposure recapitulated ALS-associated degeneration, and that we could rescue this degeneration via MAP4K4 inhibition (MAP4K4i). We subsequently identified mechanisms underlying MAP4K4i-mediated protection by performing phosphoproteomics on iPSC-derived MNs treated with ER stressors ±MAP4K4i. Through these analyses, we found JNK, PKC, and BRAF to be differentially modulated in MAP4K4i-protected MNs, and that inhibitors to these proteins could also rescue MN toxicity. Collectively, this study highlights the value of utilizing ER stressors in ALS patient MNs to identify novel druggable targets

    Filling the Gaps in the Kirromycin Biosynthesis: Deciphering the Role of Genes Involved in Ethylmalonyl-CoA Supply and Tailoring Reactions

    Kirromycin is the main product of the soil-dwelling Streptomyces collinus TĂŒ 365. The elucidation of the biosynthetic pathway revealed that the antibiotic is synthesised via a unique combination of trans-/cis-AT type I polyketide synthases and non-ribosomal peptide synthetases (PKS I/NRPS). This was the first example of an assembly line integrating the three biosynthetic principles in one pathway. However, information about other enzymes involved in kirromycin biosynthesis remained scarce.\ua0In this study, genes encoding tailoring enzymes KirM, KirHVI, KirOI, and KirOII, and the putative crotonyl-CoA reductase/carboxylase KirN were deleted, complemented, and the emerged products analysed by HPLC-HRMS and MS/MS. Derivatives were identified in mutants ΔkirM, ΔkirHVI, ΔkirOI, and ΔkirOII. The products of ΔkirOI, ΔkirOII, and kirHVI were subjected to 2D-NMR for structure elucidation. Our results enabled functional assignment of those enzymes, demonstrating their involvement in kirromycin tailoring. In the ΔkirN mutant, the production of kirromycin was significantly decreased. The obtained data enabled us to clarify the putative roles of the studied enzymes, ultimately allowing us to fill many of the missing gaps in the biosynthesis of the complex antibiotic. Furthermore, this collection of mutants can serve as a toolbox for generation of new kirromycins
